There is no permeabilization step and therefore no optimization required.

No fixation or permeabilization is required. This is one of the major advantages of Curio Seeker because that means you do not need to optimize the assay (permeabilization) for different tissues or species. Curio Seeker works on any species or tissue without the need for changes to the protocol.
