What modifications do I need to make for running Curio Seeker on plant tissue?

Currently, for plant tissues, we are not aware of anything that has to be modified. The following tips may increase the chances of success.

 

1. During the tissue dissociation step, make sure that it is done soon after sectioning (e.g. 1-2 hours after without freezing the tile). Freezing the tile and tissue (between steps 12 and 13 of Section B on the protocol) and keeping it at -80C for a couple of days may make it very difficult to dissociate.

2. Incubate the tile with tissue clearing reagents for 60 min instead of 30.
